    采用單細(xì)胞生長流動成像系統(tǒng),觀測并探究銅綠假單胞菌單細(xì)胞生長規(guī)律,運(yùn)用隨機(jī)建模的方法構(gòu)建單細(xì)胞與群體細(xì)胞之間的關(guān)系,模擬銅綠假單胞菌群體細(xì)胞的生長并進(jìn)行驗證。同時,通過多次模擬實驗來探究不同初始接菌量對銅綠假單胞菌細(xì)胞生長遲滯期的影響。結(jié)果表明,在25℃和35℃條件下,模擬曲線基本能夠呈現(xiàn)實際測得群體細(xì)胞生長的趨勢,模擬方法可以用來實現(xiàn)對群體細(xì)胞生長的預(yù)測。同時,對初始接菌量為0、1、2、4、8、16和100個/mL的銅綠假單胞菌生長進(jìn)行多次模擬表明,在25℃和35℃下,其生長遲滯時間隨初始接菌量的增大分別由12.52h降低為6.33h,由8.61h降低為4.01h。通過單細(xì)胞水平模擬群體細(xì)胞生長過程,能夠更加明顯地體現(xiàn)出細(xì)菌細(xì)胞生長過程中的不確定性與變異性,為深入研究細(xì)菌細(xì)胞生長遲滯現(xiàn)象以及準(zhǔn)確預(yù)測微生物的生長提供借鑒。

    Abstract:

    A flow chamber image system was used to study the single cell growth of Pseudomonas Aeruginosa. A stochastic modeling process was applied to connect the growth of P. Aeruginosa single cells and cell colonies, and made it possible to simulate the population growth of P. Aeruginosa. Experimental bacteria growth viable counts using different initial inoculum sizes were detected to validate the simulation process. Meanwhile, many simulations were carried out to verify the effect of inoculum size on the lag time of P. Aeruginosa. The results indicate that the agreement between simulations and viable counts is good at both 25℃ and 35℃ and the simulation process can be one of methods for predicting the population growth of P. Aeruginosa. Through many simulations, it demonstrates that the lag time decreased from 12.52h to 6.33h at 25℃ and 8.61h to 4.01h at 35℃ as the initial inoculum size increased from 0CFU/mL to 100CFU/mL. The uncertainty and variability in bacteria cell growth are shown clearly in the simulation process. Also this method can be used as a reference to predict bacterial population growth and to study the lag behavior of microorganism.
